Roles of syndecan-1 in endometrial carcinogenesis

Syndecan-1 is one of the four-membered transmembrane heparan sulfate proteoglycans (HSPG). It is well documented that syndecan-1 acts as a receptor for a wide pool of ligands and promotes growth of various cancers such as multiple myeloma, breast, and ovarian cancers. Very little, however, is known about the role of syndecan-1 associated with tumorigenesis of human endometrial cancer. So I attempted to gain insight into the correlation between expression level of syndecan-1 and its tumor-promoting characteristics in well-defined human endometrial cancer cell lines. The innate expression level of syndecan-1 was moderate to high in all 3 endometrial cancer cell lines: HEC-1A cells, KLE cells, and AN3CA cells, each representing moderately differentiated and poorly differentiated, respectively. Overexpression of syndecan-1 promoted the cancer growth by enhancing activation of both Erk and Akt. And in syndecan-1-overexpressing cells, the cytoplasmic NF-κB p65 protein is downregulated but accumulated in the nucleus. In the co-culture with normal endometrial stromal cells, overexprssion of syndecan-1 enhanced cell invasion, Integrin αv/β5 expression, and phosphorylation of FAK compared with mock transfectants. These results provide evidence that supports that syndecan-1 is associated with endometrial carcinogenesis by helping the cancer cells proliferate and propagate in the context of extracellular matrix

초록/요약

Syndecan-1은 heparan sulfate 잔기를 갖고 막을 투과하는 Proteoglycan Family의 4가지 구성원 중 하나이다. Syndecan-1은 수많은 세포 밖 리간드들의 수용기로 작용하며, 다발성 골수종, 유방암, 난소암에서 높은 발현을 보여 이들 암의 성장과 관련됨이 밝혀졌다. 하지만 사람의 자궁 내막암 발생과의 관련성은 알려진 것이 많지 않다. Syndecan-1은 분화 단계가 다른 3가지 자궁 내막암 세포주 (HEC-1A cell, KLE cell, AN3CA cell)에서 모두 높게 발현되었다. 또한 Syndecan-1을 암세포주에 과발현 시켰을 때 세포의 성장과 생존에 중요한 역할을 하는 Akt와 Erk 신호 전달 경로가 활성화되었다. 또한 과발현 시 NF-κB subunit인 p65와 p50 단백질의 세포질 양은 줄고, 핵 속으로 이동이 증가하였다. 기질 세포와의 공배양에서 Syndecan-1이 많이 발현될수록 암세포의 침윤이 증가하였다. Syndecan-1의 발현양이 늘었을 때 Integrin αV, β5 단백질의 양이 늘고, FAK 단백질의 인산화가 증가한 것으로 보아 Syndecan-1은 이들을 조절함을 알 수 있었다. 이러한 증거를 바탕으로 Syndecan-1이 자궁 내막암 세포의 성장과 전이에 밀접하게 관여함을 알 수 있다.
