DNA double-strand breaks (DSBs) can promote the formation of liquid–liquid phase separation (LLPS)-driven condensates in cells. However, the biological functions of DSB-induced condensates remain incompletely understood. In this study, I investigated the functional role of condensates formed in response to DSBs. DSB-induced condensates were formed when DSBs occurred within transcriptionally active chromatin during G1 phase, and accumulated HR-associated factors, including EXO1, BLM, and RAD51, upon entry into S/G2 phase. In addition, R-loop accumulation at DSBs was required for condensate formation, whereas SETX-dependent R-loop resolution was associated with proper condensate dissolution. When condensate formation was impaired, chromosomal instability was induced, accompanied by abnormal RAD51 distribution along tetrad-like chromosome structures. Furthermore, defective condensate formation compromised DNA replication fork integrity and reduced replication speed. Taken together, this study suggests that DSBs arising within transcriptionally active chromatin during G1 phase form condensates through R-loop accumulation, and that subsequent condensate dissolution upon S/G2 entry is accompanied by the recruitment of HR-associated repair factors, including EXO1, BLM, and RAD51, thereby contributing to the maintenance of chromosomal stability and faithful DNA replication.

DNA 이중가닥 절단(DNA double-strand breaks, DSBs)은 유전체 안정성을 위협하는 가장 치명적인 형태의 손상으로, 그 복구 과정은 세포주기 단계와 크로마틴 구조에 따라 정교하게 조절되어야 한다. 본 연구는 G1 기에 전사적으로 활성화된 영역에서 발생한 DSB 가 액체-액체 상분리(liquid–liquid phase separation, LLPS) 기반의 응집체(condensate)로 포획되며, 이 구조가 상동재조합(homologous recombination, HR)의 준비 및 진행을 조율하는 핵심 플랫폼으로 기능한다는 새로운 기전을 제시한다. 본 연구에서 확인된 응집체는 CtIP, EXO1, BLM, RAD51 과 같은 HR 관련인자를 선택적으로 농축하는 고도로 조직화된 핵 내 미세구조였으며, 이는 G1 기에서도 HR 의 초기 단계가 특정 조건에서 개시될 수 있음을 시사한다. 또한 CtIP 결손 시 응집체 해체가 지연되는 현상을 통해 응집체의 소멸이 HR 초기 이벤트와 연결되어 있음을 확인했다. 응집체 형성은 전사 활성과 크로마틴 상태 변화가 함께 관여하는 복합적인 과정으로 이루어진다. 응집체 형성은 R-loop 의 축적이 필수적이고, H3K9me3, HP1α, SUV39H1 과 같은 크로마틴 억제 인자들 역시 응집체 내부에서 검출된다. 이러한 전사 관련 요소와 억제적 크로마틴 특성은 응집체의 형성 및 안정성과 연관되어 있으며, 이들 요소 간의 정확한 시간적·인과적 관계는 아직 명확하지 않다. 그럼에도 불구하고, 이러한 특성들의 결합은 응집체가 특정 유전체 위치에서 선택적으로 형성되는 데 기여하는 것으로 보인다. 응집체의 기능적 중요성을 평가하기 위해 응집체 형성을 sorbitol 처리 또는 siCTCF 를 이용해 억제하자, 복제 스트레스 증가, S 기 진입 지연, 복제포크 속도 감소, 포크 비대칭성 증가, tetrad 유사 염색체 및 염색체 장애 등이 발생하였다. 이러한 표현형은 응집체가 단순한 구조물이 아니라, 손상된 DNA 를 안정적으로 유지하고 HR 관련 중간체의 잘못된 확산을 방지하며, 복구 완료 후 복제가 정상적으로 재개되도록 하는 기능적 관문으로 작동함을 의미한다. 특히 RAD51 이 비정상적 염색체 구조에 축적되는 현상은 응집체가 정상적인 homology search 및 strand processing 을 조절하는 데 중요한 역할을 한다는 점을 뒷받침한다. 본 연구는 LLPS, 전사, 크로마틴 재구성, DNA 손상 반응을 연결하는 새로운 개념적 모델을 확립한다. 즉, G1 기 전사활성 유전자에서 발생한 DSB 는 R-loop ·CTCF·크로마틴 리모델러의 협력으로 응집체에 포획되고, 이 구조가 HR 준비를 돕고 S 기 진입을 조절하며 복제 스트레스를 방지함으로써 유전체 안정성을 유지한다는 모델이다. 마지막으로 본 연구는 응집체 내부에서 HR 이 실제로 어느 수준까지 진행되는지, 어떤 손상 부위가 응집체를 형성할 자격을 갖는지, 응집체 해체를 유도하는 분자적 신호가 무엇인지 등 향후 탐구가 필요한 중요한 질문들을 제시한다. 이러한 한계에도 불구하고, 본 연구는 상분리 응집체가 유전체 유지를 시간적·공간적으로 정밀하게 조율하는 새로운 세포 내 기구로 작용함을 제안하며, DNA 손상 복구 연구를 넘어 전사, 크로마틴 조절, 그리고 세포주기 조절 전반에 걸쳐 중요한 통찰을 제공한다.
