Orthogonal Analytical Evidence Supports Polystyrene Biodegradation by Bacillus sp. PSNB_3001 under Defined Culture Conditions

Polystyrene (PS) is a synthetic polymer that persists extensively in the environment due to its high physicochemical stability and resistance to biodegradation. Although microbial PS degradation has been increasingly investigated, many previous studies have claimed degradability based primarily on micro- or nano-scale alterations, such as superficial surface changes or minor mass loss. However, these indirect metrics fail to exclude the confounding possibilities of abiotic physical fragmentation or surface-associated artifacts, making it fundamentally difficult to verify actual covalent polymer backbone cleavage. To address these systemic limitations, three potential PS degrading strains were isolated from compost to provide definitive evidence of macro-scale degradation by demonstrating substantial and quantifiable molecular weight reduction. Among the isolates, Bacillus sp. strain PSNB_3001 exhibited the highest activity and was selected as the primary model strain. Based on genomic analysis, an incomplete biosynthetic pathway for several amino acids was predicted and experimentally validated through amino acid omission assays. Consequently, a reduced-background defined minimal medium supplemented only with selected amino acids was developed, effectively eliminating the non-specific background growth effects commonly associated with complex media and ensuring that microbial responses were strictly driven by the polymeric substrate. Under these rigorously controlled conditions, PS incubated with Bacillus sp. PSNB_3001 underwent distinct molecular and structural transformations within 14 days. Size exclusion chromatography (SEC) revealed maximum decreases of 33.15% in weight-average molecular weight (Mw) and 81.38% in number-average molecular weight (Mn), accompanied by shifts in molecular weight distribution and increased dispersity, which directly substantiates polymer chain scission. These changes were consistent with gravimetric reduction, surface morphological alterations, and the emergence of newly detectable low molecular weight compounds verified via 1H NMR and TLC analyses. Furthermore, cell free extracellular protein fractions from Bacillus sp. PSNB_3001 induced a decrease in turbidity during PS emulsion assays, confirming the involvement of extracellular enzymes in the initial stages of PS degradation. In contrast, control groups lacking the relevant proteins showed no reduction in turbidity, indicating that the observed PS degradation was driven by genuine biological activity. While complete mineralization or metabolic assimilation of PS was not demonstrated in this study, the collective multi-analytical evidence robustly proves substantial macro-scale PS degradation by Bacillus sp. PSNB_3001. Overall, this study addresses the limitations of previous literature by providing a highly reliable, integrated analytical framework for evaluating microbial plastic degradation and proposes Bacillus sp. PSNB_3001 as a promising model strain for future investigations into polymer-microbe interaction mechanisms.

폴리스타이렌(PS)은 높은 화학적 안정성과 생분해 저항성으로 인해 환경 내에 장기 축적되는 대표적인 난분해성 플라스틱이다. 최근 미생물을 이용한 PS 분해 연구가 활발히 진행되고 있으나, 상당수 연구가 표면 변형이나 미미한 질량 감소 등 마이크로·나노 스케일(micro/nano-scale)의 간접적 지표에만 의존하고 있다. 이러한 방식은 미생물의 실질적인 대사 작용이 아닌 비생물학적 요인에 의한 물리적 파편화(ablation)나 표면 오염물 접착 등의 오인 가능성을 배제하기 어려우며, 실제 고분자 사슬의 절단 여부를 명확히 판정하는 데 근본적인 한계가 있었다. 본 연구에서는 기존 PS 분해 논문들의 이러한 고질적인 한계를 극복하고자, 퇴비로부터 잠재적 PS 분해능을 가진 균주 3종을 분리하였으며, 미세한 표면 변화가 아닌 거시적 스케일(macro-scale)에서의 실질적이고 정량적인 분자량 감소 수치를 제시하여 실제 생분해 현상을 입증하고자 하였다. 분리 균주 중 분해 활성이 가장 우수한 Bacillus sp. PSNB_3001을 모델 균주로 선정하고, 유전체 분석(Genome-guided)을 바탕으로 정밀한 PS 분해 평가 시스템을 구축하였다. 유전체 상에서 아미노산 생합성 경로의 불완전성을 예측한 뒤 이를 결핍 실험으로 검증하였으며, 이 결과를 기반으로 필수 아미노산만 선택적으로 보충한 '영양소 제한 최소배지(Reduced-background minimal medium)'를 개발하였다. 이를 통해 복합배지 사용 시 나타날 수 있는 비특이적 성장 효과를 철저히 배제함으로써, 고분자 기질 자체에 기인한 미생물 반응만을 순수하게 평가할 수 있는 기반을 마련하였다. 이처럼 철저히 통제된 조건에서 PSNB_3001을 PS와 함께 14일간 배양한 결과, 뚜렷한 분자적·구조적 변화가 관찰되었다. 크기 배제 크로마토그래피(SEC) 분석 결과, 중량평균 분자량(Mw)은 최대 33.15%, 수 평균 분자량(Mn)은 최대 81.38% 감소하였다. 이와 함께 분자량 분포의 이동과 분산도(Dispersity)의 증가가 확인되어 고분자 사슬 절단(Polymer chain scission)이 일어났음을 물리적으로 입증하였다. 이러한 거동은 실질적인 중량 감소 및 표면 형태 변형 뿐만 아니라, 1H NMR과 TLC 분석을 통해 확인된 새로운 저분자 대사산물의 생성 결과 와도 완벽히 일치하였다. 나아가 세포 외 단백질 분획을 이용한 PS 이멀젼 분석에서 뚜렷한 탁도 감소가 유도됨을 확인하여, 세포 외 효소가 PS 분해의 초기 단계에 관여함을 입증하였다. 반면 관련 단백질을 처리하지 않은 대조군에서는 탁도 변화가 나타나지 않아, 본 연구에서 관찰된 분해 현상이 순수한 생물학적 작용에 의한 것임을 확고히 뒷받침하였다. 비록 본 연구에서 PS의 완전한 무기화(Mineralization)나 대사 동화 경로까지 전면 규명하지는 못했으나, 다각적인 직교 분석(Orthogonal analysis)을 통해 PSNB_3001에 의한 거시적 수준의 고분자 절단 현상을 성공적으로 증명하였다. 결론적으로 본 연구는 기존 문헌들의 한계를 보완하여 미생물의 플라스틱 분해 성능을 신뢰성 있게 평가할 수 있는 통합 분석 프레임워크를 제시하며, PSNB_3001이 향후 난분해성 고분자와 미생물 간의 상호작용 기작을 규명하기 위한 유망한 모델 균주가 될 수 있음을 시사한다.
