Structure-based discovery of TI-16 as a small-molecule modulator of TNF-α– TNFR1 interaction and inflammation

Tumor necrosis factor-alpha (TNF-α) is a central mediator of inflammatory diseases and contributes to progressive tissue damage through the induction of apoptotic and necroptotic cell death, as well as the dysregulated production of pro-inflammatory cytokines. Although TNF-α-targeting monoclonal antibodies have demonstrated significant clinical efficacy, their widespread use is limited by several drawbacks, including the requirement for parenteral administration, immunogenicity, and high treatment costs. Consequently, considerable efforts have been directed toward the development of orally available small-molecule TNF-α inhibitors as a more accessible and cost-effective therapeutic alternative. However, despite the pivotal role of TNF-α in chronic inflammatory disorders, no small-molecule TNF-α inhibitor has yet received clinical approval, and only a limited number of candidates have entered clinical development. To identify novel TNF-α inhibitors, we performed structure-based virtual screening of chemical libraries, followed by comprehensive in vitro validation assays to evaluate their biological activities and therapeutic potential. Through this approach, we identified TI-16 as a potent small-molecule inhibitor of TNF-α. TI-16 effectively protected human and mouse cells from TNF-α-mediated cytotoxicity and significantly reduced inflammatory responses. Notably, TI-16 suppressed both apoptotic and necroptotic signaling pathways, indicating its ability to broadly inhibit TNF-α-driven cell death programs. Mechanistic studies involving immunoblotting, biophysical analyses, and molecular dynamics simulations revealed that TI-16 selectively disrupts TNF-α–TNFR1 binding without affecting TNF-α trimerization. By targeting the initial ligand–receptor interaction, TI-16 effectively suppressed a wide range of TNFα-mediated downstream signaling events. Collectively, these findings demonstrate that TI-16 possesses a unique mechanism of action, broad anti-inflammatory efficacy, and potential suitability for oral administration. Therefore, TI-16 represents a viable lead compound for the development of TNF-α-targeted therapeutics that may address the limitations associated with currently available biologic agents.

종양괴사인자-알파(TNF-α)는 염증성 질환의 핵심 매개 인자로서, 세포사멸(apoptosis) 및 괴사성 세포사멸(necroptosis)을 유도하고 염증성 사이토카인의 비정상적인 분비를 촉진함으로써 지속적인 조직 손상에 기여합니다. TNF-α를 표적으로 하는 단일클론 항체 치료제는 우수한 임상적 효능을 입증하였으나, 주사 투여의 필요성, 면역원성 및 높은 치료 비용 등의 한계로 인해 사용에 제약이 있습니다. 하여, 현재 시장에서는 보다 접근성이 높고 경제적인 치료 전략으로서 경구 투여가 가능한 저분자 TNF-α 억제제 개발에 대한 관심이 증가하고 있습니다. 그러나 TNF-α가 만성 염증성 질환의 병태생리에 핵심적인 역할을 수행함에도 불구하고, 현재까지 임상 승인을 받은 저분자 TNF-α 억제제는 없으며, 소수의 후보 물질만이 임상 개발 단계에 진입한 상태입니다. 본 연구에서는 새로운 TNF-α 억제제를 발굴하기 위하여 화합물 라이브러리를 대상으로 구조 기반 가상 스크리닝(structure-based virtual screening)을 수행하였으며, 이후 다양한 in vitro 검증 실험을 통해 생물학적 활성 및 치료 가능성을 평가하였습니다. 그 결과, 우리는 TI-16을 TNF-α에 대한 강력한 저분자 억제제로 규명하였습니다. TI-16은 TNF-α에 의해 유도되는 세포독성으로부터 인간 및 마우스 세포를 효과적으로 보호하였으며, 염증 반응을 유의하게 감소시켰습니다. 특히 TI-16은 세포사멸 및 괴사성 세포사멸 신호전달 경로를 모두 억제함으로써 TNF-α에 의해 유도되는 광범위한 세포사멸 프로그램을 효과적으로 차단할 수 있음을 보여주었습니다. 면역블롯 분석, 생물물리학적 분석 및 분자동역학 시뮬레이션을 포함한 기전 연구를 통해 TI16은 TNF-α의 삼량체 형성에는 영향을 미치지 않으면서 TNF-α와 TNFR1 간의 상호작용을 선택적으로 저해하는 것으로 확인되었습니다. 이러한 결과는 TI16이 ligand-receptor 결합의 초기 단계를 표적으로 함으로써 TNF-α 매개 하위 신호전달 경로를 광범위하게 억제할 수 있음을 시사합니다. 종합적으로, 본 연구 결과는 TI-16이 독창적인 작용 기전과 우수한 항염증 효능을 보유하고 있으며 경구 투여형 치료제로 개발될 가능성을 가진 유망한 후보 물질임을 보여주고 있습니다. 따라서 TI-16은 현재 사용되고 있는 생물학적 제제의 주요 한계를 극복할 수 있는 새로운 TNF-α 표적 치료제 계열 개발을 위한 유망한 선도물질(lead compound)로 평가됩니다.
