초록/요약
본 연구는 돈피 유래 아텔로콜라겐 (Porcine derived Atelo Collagen, PSAC)의 정제 공정을 최적화하고, 제조된 시료의 이화학적 특성과 in vitro 및 in vivo 효능을 평가하는 것을 목적으로 하였다. 제조 공정은 전 공정을 4℃에서 유지하면서, 규조토를 배제한 다단계 심층 여과와 산성 조건의 TFF 탈염 공정을 중심으로 설계하였다. 최종적으로 제조된 PSAC 는 수율, 단백질 함량, 콜라겐 순도, 구조적 특성, 생체적합성 및 동물 모델에서의 효능을 기준으로 평가되었다. 연구 결과, 원료 90 g 에서 공정을 시작하여 최종적으로 동결건조 시트로서, 약 2.3 g 의 아텔로콜라겐을 회수하여 약 2.6%의 수율을 확보하였다. TFF 탈염 공정에서는 전도도가 초기 121 ms/cm 에서 diafiltration 5 cycle 후 5.5 mS/cm 수준으로 감소하여 공정 종결점을 정량적으로 설정할 수 있었다. 또한 규조토를 사용한 경우 단백질 회수율이 0.2% 수준에 불과한 반면, 규조토를 배제한 경우 회수율이 93.4%로 크게 향상되어 다단계 심층 여과의 유효성이 확인되었다. 제조된 시료의 이화학적 특성을 평가하기 위해 단백질 함량 분석 결과, PSAC 의 보정 단백질 농도는 945.92±0.69 µg/mL 로 reference collagen 과 동등한 수준을 보였으며, 하이드록시프롤린 분석을 통해 콜라겐 함량이 95.5%로 확인되었다. FT-IR 분석에서는 Amide A, I, II, III 영역에서 reference collagen 과 유사한 피크 위치를 나타내어 콜라겐 고유의 삼중나선 구조가 유지됨을 확인하였다. 또한 WST-1 assay 에서 HDFn 세포에 대한 세포생존율이 모든 농도에서 100% 이상으로 나타나, 공정 중 사용된 산 및 펩신 등의 잔류 독성이 효과적으로 제거되었음을 확인하였다. 동물 효능 평가에서는 악력, 인장강도, 운동성 및 H&E 조직학적 평가를 수행하였다. 그 결과, PSAC 처리군은 기능적 회복과 조직 재생 측면에서 긍정적인 경향을 나타냈으며, 특히 인장강도는 2 주차에 정상군의 약 96% 수준에 도달하였다. 조직학적으로는 4 주차에 손상 부위 간극 감소와 이식재의 생체 내 융화 양상이 관찰되었다. 본 연구에서 확립한 정제 공정이 고순도 및 안정성이 확보된 돈피 유래 아텔로콜라겐 제조에 적합하며, 향후 의료용 생체재료로의 응용 가능성이 있음을 시사한다.

This study aimed to optimize the purification process of porcine skin-derived atelocollagen (PSAC)and evaluate its physicochemical properties along with its in vitro and in vivo efficacy. The manufacturing process was designed to be maintained at 4°C throughout all stages, focusing on multi-stage depth filtration excluding diatomaceous earth and a tangential flow filtration(TFF)based desalting step under acidic conditions. The finalized PSAC was evaluated based on yield, protein content, collagen purity, structural characteristics, biocompatibility, and therapeutic efficacy in an animal model. As a result, approximately 2.3 g of atelocollagen was recovered from 90 g of raw porcine skin, securing a yield of approximately 2.6% based on the final lyophilized sheet type. In the TFF desalting process, the conductivity decreased from an initial 121 mS/cm to 5.5 mS/cm after 5 diafiltration cycles, allowing the process endpoint to be established quantitatively. Furthermore, while the protein recovery was only 0.2% when diatomaceous earth was used, it significantly improved to 93.4% when diatomaceous earth was excluded, confirming the high efficiency of the sequential multi-stage depth filtration. To evaluate the physicochemical properties of the manufactured samples, total protein content analysis was conducted. The corrected protein concentration of PSAC was 945.92±0.69 µg/mL, demonstrating a level comparable to that of reference collagen, and hydroxyproline analysis confirmed a collagen purity of 95.5%. FT-IR analysis revealed peak positions in the Amide A, I, II, and III regions similar to those of the reference collagen, indicating that the native triple-helical structure of collagen was well-preserved. Moreover, WST-1 assay demonstrated a cell viability of over 100% against HDFn cells at all tested concentrations, confirming that residual toxic materials such as acids and pepsin used during the process were effectively removed. In the animal efficacy study, grip strength, tensile strength, motility, and H&E histological evaluations were performed. The PSAC-treated group exhibited favorable trends in functional recovery and tissue regeneration; notably, the tensile strength reached approximately 96% of the normal group at week 2. Histologically, narrowing of the damaged tissue gap and in vivo integration of the implant were observed at week 4. In conclusion, the optimized purification process established in this study is highly suitable for manufacturing porcine skin-derived atelocollagen with high purity and stability, suggesting its strong potential for future applications as a medical biomaterial for rotator cuff disease.
