A Study on the Skin-Barrier-Enhancing, Acne- Impoving, and moisturizing Effects of Organic Plant Extract Complex Graduate School of Ajou University Department of Pharmacy Chang Kee Jong With the increasing prevalence of skin sensitivity and environmental stressors, there is a growing demand for natural and organic cosmetic ingredients that offer both high functionality and safety. In this study, we developed a liposomal formulation containing a complex of five organic plant extracts—Camellia sinensis, Chamomilla recutita, Centella asiatica, Aloe barbadensis, and Lavandula angustifolia—and evaluated its moisturizing, anti-inflammatory, barrier- strengthening, and sebum-regulating effects through a series of in vitro and in vivo studies. In vitro, the liposomal complex significantly suppressed nitric oxide (NO) production by 31% in LPS-stimulated cells, enhanced hyaluronic acid synthesis by 62%, and markedly upregulated the expression of barrier-related proteins, including filaggrin (3.4-fold) and involucrin (2.3-fold), compared with untreated controls. In vivo, a tape-stripping model was used to induce transient skin barrier disruption. Thirty minutes after application, the treated sites exhibited a significantly higher transepidermal water loss (TEWL) recovery rate (71.1%, p<0.001) and improved erythema index (71.5 A.U., p<0.001) compared with untreated sites. Cumulative irritation testing demonstrated a mean irritation score of 0.40, classifying the formulation as “low irritancy.” Furthermore, a 4-week clinical study demonstrated a significant reduction in sebum excretion (41%, p<0.001), accompanied by decreases in non-inflammatory (23.3%) and inflammatory (25.8%) acne lesion counts (p<0.01 for both), with no adverse events reported throughout the study period. Taken together, these findings indicate that the Organic MicroMelt Complex Calming liposomal formulation effectively reinforces the skin barrier, enhances hydration, suppresses inflammatory responses, and regulates sebum secretion, while maintaining an excellent safety profile. This formulation shows strong potential as a next-generation functional ingredient for organic cosmetics, particularly for sensitive and acne-prone skin.
