The anti-inflammatory effects of Aiouea padiformis extract via NLRP3 inflammasome inhibition

초록/요약
Aiouea padiformis, a member of the Lauraceae family found in tropical regions, has not been well-studied for its anti-inflammatory effects, despite reports of similar properties in other Aiouea species. In this study, I investigated the anti-inflammatory potential of Aiouea padiformis extract (AP) and its underlying mechanisms, focusing on the NOD-, LRR-, and pyrin domain-containing protein 3 (NLRP3) inflammasome. AP treatment inhibits NLRP3 inflammasome-mediated interleukin-1 beta (IL-1β) secretion in J774A.1 and THP-1 cells without affecting cell viability. Moreover, AP did not alter upstream events associated with NLRP3 inflammasome activation, such as NF-κB signaling, potassium efflux, or intracellular reactive oxygen species (ROS) production. Interestingly, AP significantly reduced the ATPase activity of NLRP3, leading to the inhibition of ASC oligomerization and speck formation. These results suggest that AP holds promise as a natural anti-inflammatory agent targeting NLRP3 inflammasome activity.
