젊거나 노화된 멜라노사이트에서 분비하는 엑소좀의 생화학적 특성 연구

초록/요약

노화와 관련하여 aged melanocyte와 young melanocyte에서 분비되는 exosome을 연구하였다. Melanocyte에서 exosome이 cell의 어느 위치에 존재하는가를 형광물질을 표지하고 confocal microscopy를 이용하여 확인하였는데 cell의 중심부가 아닌 바깥쪽에 위치하는 것을 확인하였다. Exosome의 크기를 측정하여 배지 속에 다른 종류의 extracellular vesicles 중에서 exosome으로 추정되는 크기 범위에 입자들을 측정하였다. 측정 결과 exosome으로 생각 되는 30 ~ 150 nm사이의 입자들이 측정되었다. Western blot을 이용하여 exosome의 marker protein인 CD81과 fibronectin을 확인하였다. CD81은 CD9, CD63 등과 같은 tetraspanin로 exosome에서 가장 많이 확인하는 단백질이다. Silver staining을 통해서 aged melanocyte와 young melanocyte에서 분비된 단백질의 양이나 종류에 관한 차이점을 확인하였다. Gel안에서 발현 차이점이 있는 단백질을 분석하기 위해서 mass spectrometry를 수행하였고 세포 내 중요한 인자들인 cell cycle, chaperone, growth factor 등을 기준으로 몇 가지 단백질을 정하여 비교하였다
