Studies on skin moisturizing and anti-inflammatory efficacy of peptide and amino acid hydrolysate from ginseng seed

The aim of this study was to determine skin moisturizing and anti-inflammatory efficacy of complex peptide & amino acid hydrolysate derived from ginseng seed protein. It’s believed that ginseng seeds contain a variety of functional and bioactive protein. Ginseng seed protein was extracted 18.9% of yields from defatted Korean Panax ginseng C.A. Meyer seeds. In order to solve transdermal problem of protein and enhance skin penetration, then the protein was digested by a parallel combination of bromelain and exo-peptidase as protease to produce small molecular peptides and amino acids in the hydrolysate. They are easy to penetrate into skin. The resultant hydrolysate was comprised by 36% amino acids and around 60% small peptide (<1000Da). After purifying, the hydrolysate from ginseng seed protein was investigated on efficacy test of skin moisturizing and anti-inflammation. The study showed moisture contents in stratum corneum(SC) increased around 20% over 5hrs of sample application. NMFs contents increased 51% and lipid contents increase 20% more than control in SC. Furthermore, proteomic SDS-PAGE analysis of HaCaT cells indicated complex peptide and amino acid hydrolysate up-regulated expression of the protein related to 3 moisturizing mechanisms, such as JAM-1 upped to 831%, involucrin upped to 1573%, AQP-3 upped to 519%. In addition, the 3000ppm of complex peptide and amino acid hydrolysate inhibited 60% nitric oxide (NO) production from LPS-induced RAW 264.7 cell. In the further study, some key inflammatory “messenger” protein in the RAW 264.7 cell had significant down regulation because of the complex hydrolysate. Such as TLR4 downed to 35%, IRAK4 and Ikkα downed to 15.2% and 4.1% respectively et al. The results demonstrated complex peptide & amino acid hydrolysate from ginseng seed protein is a potential active ingredient that can be applied for skin moisturizing and anti-inflammation efficacy in the cosmetics.
