Boronate-diol 작용에 의한 HbA1c의 수평 이동 발색 측청법에 관한 연구

A lateral-flow chromogenic assay of human HbA1c based on the boronate-diol interaction

초록/요약

인체 혈액에 존재하는 HbA1c 농도는 지난 수 개월 동안의 평균 혈당을 반영하는 것으로 알려져 있다. 그리하여, HbA1c 수준의 측정은 당뇨병 환자들의 임상 관리에서 필수적인 요소로 작용하고 있다. 특별히 HbA1c를 분석하기 위해 azo-BA dye를 사용하여 glycated hemoglobin(HbA1c)에 대한 확인 방법을 고안해냈다. 본 연구에서 lateral-flow 방식의 발색측정법으로 nitrocellulose membrane을 사용해 실제 biosensing에 적용시켜 사람의 HbA1c 샘플을 가지고 HbA1c 분석 연구가 진행된다. 용혈된 혈액 내 존재하는 total hemoglobin은 azo-BA dye와 섞인 다음에 glycosylated hemoglobin(HbA1c) 부분이 cis-diol interaction을 통해 azo-BA dye의 boronate group에 특이적으로 결합하게 된다. 첫째로, HbA1c/Azo-BA dye 화합물이 membrane 상 특정 부위에 주입된 후 azo-BA dye의 boronate group에 HbA1c가 농도 경향성이 나타나도록 결합하게 한 다음, 버퍼를 흘려준 뒤 HbA1c/Azo-BA dye 화합물이 membrane 상의 zinc가 있는 부위로 옮겨가도록 해준다. Zinc ion을 사용하여 HbA1c/Azo-BA dye 화합물이 membrane상에 zinc가 있는 부위에 결합되도록 한다. 그래서 membrane상에 zinc가 있는 부위에 HbA1c의 퍼센타일 농도에 따라서 오렌지색에서 붉은 자주색으로 변하는 azo-BA dye 색의 변화와 azo-BA dye의 boronate group에 HbA1c가 결합하는 것을 관찰할 수가 있다. 간편하게 분석할 수 있고 이러한 실험방법이 point-of-care-test(POCT)에 적용되도록 HbA1c/Azo-BA dye 혼합물과 zinc ion으로 구성된 간단한 확인 방법을 마련하였다.

초록/요약

The concentration of HbA1c that is present in human blood is known to reflect the average blood glucose concentration over the past three months. Thus, the measurement of HbA1c level is an essential tool in the clinical management of diabetes mellitus. We have developed an confirming method for glycated hemoglobin (HbA1c) using Azo-BA(boronic acid)-dye to specifically analyze HbA1c. In this study, HbA1c analysis was performed with human HbA1c samples as a real biosensing application using nitrocellulose membranes as lateral-flow chromogenic assay. The hemolysed total hemoglobin is mixed with Azo-BA dye, and glycosylated hemoglobin (HbA1c) portion binds specifically to boronate groups of Azo-BA dye via cis-diol interaction. First, HbA1c /Azo-BA dye conjugates are injected on the surface of membrane strip. The concentration-dependent binding of HbA1c to boronate groups of Azo-BA dye was confirmed. Next, if buffer solution flows, HbA1c /Azo-BA dye conjugates is moving to the zinc-moified part on the surface of membrane srip. Using zinc ions, HbA1c /Azo-BA dye conjugates could be bound to the zinc-modified part on the surface of membrane strip. So we could observe the color change of Azo-BA dye from orange(before reaction of HbA1c and Azo-BA dye) to reddish purple(after reaction of HbA1c and Azo-BA dye) and the binding of HbA1c to boronate groups of Azo-BA dye, according to the percentile concentration of HbA1c, at the zinc-modified part on the surface of membrane strip. To perform the analysis with ease and to develop this analytical strategy into a point-of-care-device, a simplified confirming method consisting of a HbA1c /Azo-BA dye conjugate and Zinc ions is designed and constructed.
