the Effect of Ultrasound on Water Transport in Synovial Membrane

Purpose: The aims of this study were to investigate the effects of ultrasound on water transport and its relationship to expression of AQP-1 in synovial membrane. Materials and Methods: Enhancement of permeability for a sodium fluorescein in human synovial membrane was achieved using ultrasound at a frequency of 1MHz, intensity of 100mW/cm2 with an exposure duration of 20 minutes. To determine effects of ultrasound on AQP-1 in synoval membrane, we compared the control group, HgCl2-treated group, ultrasound-treated group and ultrasound plus HgCl2-treated group. Light microscopy and scanning, transmission electron microscopy were used to observe ultrasound-induced structural changes in synovial membrane. Also we observed the expression of AQP1 by the ultrasound stimulation with western blot analysis and immunohistochemistry. Results: The permeability was increased by ultrasound was applied at 100mW/cm2. The permeability of the US-treated synovial membrane increased about 20% when compared with control group at 6 hours after. And HgCl2 treated group, permeability decreased about 13%~15% compared to non-treated group. The ultrasound stimulus made the membrane smooth. The ultrasound group lessened fibrous degree compared to control group. And ultrasound stimulus increased the AQP1 expression in synovial membrane when examined by immunohistochemistry. Conclusion: The ultrasound stimulus increases the permeability of synovial membrane through the structural change and increase of AQP1.
