Effects of Cleyera japonica on MUC5AC expression and secretion in airway epithelial cells

Cleyera japonica is known to inhibit inflammatory cytokines and to prevent or treat various allergic diseases by reducing or neutralizing allergenic proteins derived from house dust mite. Also, Plant extracts, including Cleyera japonica, provide compositions for the prevention or treatment of asthma. Based on this fact, we investigated whether Cleyera japonica considerably affects the gene expression and secretion of airway mucin. Confluent NCI-H292 cells were pretreated with Cleyera japonica for 30 min and then stimulated PMA (phorbol 12-myristate 13-acetate) for 24 h or the indicated periods. The MUC5AC gene expression was measured by RT-PCR. Secretion of MUC5AC protein were measured by ELISA. To elucidate the action mechanism of Cleyera japonica, effect of Cleyera japonica on PMA-induced MAPK signaling pathway was investigated by western blot analysis. The results were as follows: (1) Cleyera japonica inhibited the expression of MUC5AC protein induced by PMA; (2) Cleyera japonica inhibited the secretion of MUC5AC induced by PMA; (3) Cleyera japonica inhibited PMA-induced phosphorylation of p38, ERK and JNK. This result suggests that Cleyera japonica can regulate the gene expression, secretion of mucin by acting on airway epithelial cells via regulation of MAPK signaling pathway.
